Abstract

Investigation on the influence of extra-cellular matrix components and organic detergents, on the direction of cell in vitro-differentiation from common precursors. The main goal was directed to searching and investigation, of mechanisms for activation of normal cell differentiation, with suppression of malignant cell transformation, at the same time, by development of ways for safe application, in particular, to the role of components from Cul-Nedd-, STAT-, and NF-κB-mediated cascade regulatory pathways in these processes. Additional copy of oncogene Dcn1 was inserted in normal mouse ESCs, for eventual elongation of life, it was important preserved normal/non-malignant characteristics to be tested and proved. On the other hand, additional copy of tumor-suppressor gene was inserted in malignant cells, for eventual decrease of their malignant potential. Fibroblasts from 3T3 cell line, derived from Balb/c mouse embryos, were propagated. One part of them was pre-incubated in cultural fluid, in which mouse malignant myeloma cells were previously cultivated, for a period of 2-3 days. After freezing of separate sub-populations from the so incubated cells for 2-4 weeks in the presence of cryo-protector Dymethylsulfoxide (DMSO), subsequent thawing and re-incubation, appearance of osteoclast-like cells was observed. On the other hand, signs of osteoblast lineage differentiation of from the derived of 3T3 cells, incubated separately for a long-term period in the presence of cultural fluid, supplemented after previous cultivation of malignant mouse myeloma cells in it, were noted.
